Changes and unusual items
- ARCALYST revenue increased 102% year over year in Q2 and 98% for the first half; management attributed the increase primarily to more patients.
- Q2 license and collaboration revenue included a $15.0 million Genentech development milestone for a new indication. Kiniksa recognized $14.0 million of Q2 revenue related to performance obligations satisfied in prior periods following that milestone.
- Net income was materially affected by a $19.2 million benefit from releasing the valuation allowance on U.S. deferred tax assets. The company reported an income tax benefit of $16.2 million in Q2 and $13.3 million for the first half. Management expects reported income tax expense to increase in future periods.
- R&D spending rose notably on KPL-404 clinical manufacturing and its Phase 2 rheumatoid arthritis trial; spending on mavrilimumab and vixarelimab declined from the prior-year periods.
- Operating cash use improved to $8.0 million for the first half from $44.7 million, although working-capital movements included higher receivables and lower accounts payable.
Outlook, risks and commitments
Management expects available cash, cash equivalents and short-term investments to fund operations and capital expenditure requirements for at least 12 months from issuance of the financial statements. The company nevertheless expects operating losses for the foreseeable future and may need additional capital over time; sustained viability depends on ARCALYST sales and/or financing.
- Kiniksa expects KPL-404 Phase 2 trial data in rheumatoid arthritis in the first half of 2024. Enrollment in the proof-of-concept portion was ongoing.
- The ARCALYST drug-substance manufacturing technology transfer from Regeneron to a new CDMO is a significant execution and supply risk. The filing describes a $97.3 million minimum commitment to the new CDMO, with $20.4 million due within one year. Total minimum manufacturing commitments across agreements were $139.4 million, of which $62.5 million was due within one year.
- Under the Genentech agreement, approximately $585 million of potential contingent payments remained as of June 30, 2023, before upstream obligations; these milestones are contingent, not guaranteed revenue. Kiniksa also has potential tiered royalties.
- Principal risks include reliance on ARCALYST, payer coverage and market adoption, competition, clinical and regulatory uncertainty, dependence on third-party manufacturers and CROs, and the possibility of delays or cost increases in manufacturing and development. Management reported no material litigation and no substantial doubt about the company’s ability to continue as a going concern within the assessed one-year period.
Important facts for investors to verify
- Whether ARCALYST patient growth and revenue momentum continue, and how payer coverage changes affect access and net revenue.
- Timing, cost and regulatory comparability of the ARCALYST manufacturing transfer, and whether supply remains adequate during the transition.
- Progress and results from the KPL-404 Phase 2 program and the expected first-half 2024 data timing.
- Cash consumption, working-capital trends and the company’s ability to meet substantial manufacturing commitments without additional financing.
- The sustainability of reported earnings: first-half net income depended materially on the deferred-tax valuation-allowance release, while operating results remained loss-making.
- Realization and timing of Genentech contingent milestones and royalties; the remaining milestone pool is subject to development, regulatory and sales conditions.
