Net cash used in operating activities$43.2 million$34.4 million
Cash, cash equivalents and marketable securities$885.4 million at March 31$930.9 million at December 31, 2023

As a pre-revenue company, operating and net profit margins are not meaningful. The net loss widened by $6.2 million year over year as operating expenses increased; higher interest income partly offset the increase. R&D growth reflected azetukalner manufacturing and clinical-development activity, personnel costs and stock-based compensation. G&A growth reflected higher personnel and stock-based compensation, professional fees and IT costs.

Cash and cash equivalents were $58.9 million and marketable securities totaled $826.5 million at quarter-end. Investing activities used $46.2 million, principally reflecting purchases of marketable securities net of maturities; this portfolio activity contributed to the decline in cash balances and should be distinguished from operating cash burn. Total assets were $919.0 million and shareholders’ equity was $887.7 million. Total liabilities were $31.3 million, including $10.4 million of operating lease liabilities; the filing reports no material debt balance.

Management expects existing cash, cash equivalents and marketable securities, as of the report date, to fund operating expenses and capital expenditures for at least the next 12 months. It expects continuing losses and substantial expenses for the foreseeable future and may seek funding through collaborations, equity or debt financing. The company had an at-the-market program authorizing up to $250 million in gross share sales; $29.5 million of net proceeds had been raised under it through March 31, 2024.

Business developments, outlook and risks

  • Azetukalner (XEN1101): The lead candidate is in Phase 3 development for focal onset seizures (X-TOLE2 and X-TOLE3) and primary generalized tonic-clonic seizures (X-ACKT). Management expected X-TOLE2 enrollment to finish in late 2024 to early 2025.
  • Major depressive disorder: Following the Phase 2 X-NOVA topline results reported in November 2023, management expected to present data at a May 28–31, 2024 meeting and anticipated initiating the first of three Phase 3 trials in the second half of 2024. An investigator-sponsored Phase 2 study was also ongoing.
  • Other programs: Xenon’s near-term discovery focus includes Kv7 channel openers, Nav1.7 inhibitors and Nav1.1 openers, with the goal of advancing multiple candidates into IND-enabling studies in 2024 and 2025. Neurocrine Biosciences is developing partnered candidate NBI-921352 in a Phase 2 trial.
  • Key risks: The business depends substantially on azetukalner’s clinical and regulatory success. Trials may be delayed, fail to demonstrate safety or efficacy, or require additional studies; the company also depends on third parties for manufacturing and trial conduct. Xenon has no product revenue and expects ongoing losses, and future financing may be dilutive or unavailable on acceptable terms.
  • Other exposures and contingencies: The company does not hedge Canadian-dollar exposure. It estimated that a 100-basis-point interest-rate increase would reduce marketable-securities fair value by approximately $4.8 million. Up to $6 million in regulatory milestone payments may be due to 1st Order Pharmaceuticals; no royalties are payable to 1st Order. Management reported no material legal proceedings and no material change in previously disclosed contractual commitments.

Investor verification points

  • Track enrollment and execution of the azetukalner Phase 3 epilepsy trials, and verify subsequent clinical updates against the stated timelines.
  • Review the full X-NOVA data and the planned MDD Phase 3 design and initiation timing; the filing’s outlook is forward-looking, not a guarantee.
  • Monitor operating cash use, investment maturities and the stated minimum 12-month runway, including whether plans or funding needs change.
  • Assess potential dilution from equity compensation, pre-funded warrants and any future use of the ATM program.
  • Evaluate manufacturing, trial-enrollment, regulatory, collaboration and intellectual-property risks as programs advance.
