Prothena's Partnership Strategy: Growth Driver or Limitation?

Prothena Corporation PRTA is focused on developing treatments for neurodegenerative and rare diseases. The company’s growth strategy relies significantly on partnerships with larger pharmaceutical companies to advance its drug candidates through clinical development and potentially toward commercialization. These collaborations provide financial support and access to development expertise, but they also mean that Prothena loses control over key programs.

Developing medicines requires substantial investment, and clinical trials can take several years to complete. Prothena’s partnerships help reduce the financial burden of advancing its pipeline independently. By working with established pharmaceutical companies, this clinical-stage biotechnology company can access additional research capabilities, clinical development expertise and commercial infrastructure. These arrangements may also provide milestone payments when specific development or regulatory goals are achieved, along with royalties if partnered medicines reach the market and generate sales. This approach enables the company to pursue multiple drug development opportunities without funding every development stage on its own.

However, the financial benefits depend on clinical progress, the terms of each agreement and the eventual commercial success of the partnered medicines.

RHHBY/PRTA Partnership Boosts Prasinezumab’s Development

One of Prothena’s key partnerships is with Roche RHHBY, which is developing prasinezumab for the potential treatment of Parkinson’s disease. Roche initiated the phase III PARAISO study in late 2025, with primary completion expected in 2029. The partnership allows Prothena to benefit from Roche’s clinical development and commercialization capabilities without bearing the full cost of advancing the medicine independently.

Prasinezumab could become an important source of milestone payments and royalties for Prothena if it achieves regulatory approval and generates sales. However, the drug is still investigational and its commercial potential depends on the results of ongoing clinical studies. Any disappointing results or delays could affect the timing and value of future payments to Prothena.

NVO Partnership Creates Another Growth Opportunity

Prothena’s partnership with Novo Nordisk NVO provides another potential source of long-term growth. Novo had earlier acquired Prothena’s clinical-stage antibody, Coramitug (formerly PRX004), a first-in-class amyloid depleter antibody for the treatment of ATTR amyloidosis with cardiomyopathy (ATTR-CM), a serious condition in which abnormal protein deposits can affect the heart.

Novo is studying the candidate under a late-stage CLEOPATTRA study for ATTR-CM. The study is expected to be completed by 2029. The ongoing open-label study evaluates the biodistribution of 89Zr-coramitug, and the assessment of coramitug’s ability to reduce TTR amyloid deposits in myocardial tissue using PET/CT imaging in patients with ATTR-CM is ongoing, with primary completion expected in 2027.

In March 2026, Prothena received a $50-million clinical milestone payment tied to the enrollment progress in the ongoing study of coramitug. This payment demonstrates how clinical progress in a partnered program can generate revenues for Prothena before a medicine reaches the market.

Coramitug could provide additional milestone payments and royalties if development progresses successfully. The program still faces clinical and regulatory risks and its ultimate commercial value will depend on the treatment’s safety, effectiveness and ability to compete in the ATTR-CM market.

Bristol Myers Squibb Partnership Expands PRTA’s Pipeline

Prothena’s collaboration with Bristol Myers Squibb BMY adds another potential growth driver. The latter company is evaluating moponetug, also known as BMS-986446, in the phase II TargetTau-1 study for early Alzheimer's disease. The study was fully enrolled, with primary completion expected in the first half of 2027.

The partnership gives Prothena exposure to the potential Alzheimer's treatment market without requiring it to fund the entire development program independently. If the clinical results support further development, the program could generate milestone payments and future royalties.

Bristol Myers Squibb is also evaluating PRX019 for neurodegenerative diseases. The company in-licensed exclusive global rights to PRX019 in 2024. Prothena completed a phase I study evaluating the safety, tolerability, immunogenicity and pharmacokinetics of single-ascending and multiple-dose administration in healthy volunteers.

The company could receive a potential $55-million clinical milestone payment if Bristol Myers Squibb elects to advance the program, with a decision expected by the end of 2026.

Prothena's partnered programs could provide substantial financial benefits over time. In its February 2026 business update, the company estimated that these programs could generate $3 billion in aggregate future milestone payments, in addition to potential royalties.

Prothena's partnerships with the pharma bigwigs provide access to established drug development capabilities, and potential future milestone payments and royalties.

Clinical setbacks, changes in a partner's priorities or a decision to discontinue a program could reduce the company's future revenue opportunities.
