Changes and drivers
- Revenue increased $33.9 million year over year, primarily because a $75.0 million Vertex clinical-advancement milestone was achieved in Q1. Entrada recorded a $38.7 million cumulative catch-up adjustment for that milestone; the company expected to receive the payment in Q2 2024.
- Q1 collaboration revenue included $58.1 million under the Vertex agreement and $1.0 million under the Pierrepont agreement. Vertex revenue included $2.0 million of cost reimbursements and $17.4 million recognized from opening deferred revenue.
- Vertex revenue also included a $12.2 million cumulative catch-up adjustment following revisions to estimated remaining research costs, primarily reflecting manufacturing efficiencies.
- R&D expense rose $5.5 million, mainly from higher internal personnel and facility costs and increased candidate-development costs. G&A expense rose $1.5 million, primarily from personnel costs.
- Net income improved by $30.2 million year over year, reflecting higher collaboration revenue and interest income. The 2023 operating cash inflow was largely driven by the Vertex upfront payment; Q1 2024 operating cash use reflected changes in operating assets and liabilities, including the increase in collaboration receivable.
- Marketable securities declined to $259.0 million from $284.4 million at year-end. The company reported $441,000 of unrealized losses on securities in Q1.
Outlook, risks and unusual items
- Management expects cash, cash equivalents and marketable securities at March 31, ongoing Vertex research support, and the expected $75.0 million milestone receipt to fund operations through Q2 2026. This estimate depends on assumptions that may prove incorrect. The company also stated that its available resources are expected to cover at least the next 12 months.
- Management expects operating losses and negative operating cash flows to continue as development advances; substantial additional funding will be needed over time. The $150.0 million ATM program had no share sales through March 31.
- ENTR-601-44’s U.S. IND remains on FDA clinical hold. A UK Phase 1 healthy-volunteer trial was dosing its fourth and final cohort; data were expected in October 2024. Entrada planned Phase 2-enabling regulatory submissions for ENTR-601-44 and ENTR-601-45 in Q4 2024, an ENTR-601-50 submission in 2025, and selection of an exon 51 candidate by year-end 2024.
- Vertex is responsible for global development, manufacturing and commercialization of VX-670 beyond Entrada’s research work. On May 6, 2024, after quarter-end, Vertex announced that its Phase 1/2 trial authorizations had cleared in the U.S., Canada, UK, EU and Australia, with enrollment and dosing underway.
- A $1.0 million OSIF sublicense fee related to the Q1 Vertex milestone was accrued and recorded in R&D expense. Income tax expense was $1.8 million, largely reflecting projected current tax liabilities associated with Vertex payments; the company maintained a valuation allowance on remaining deferred tax assets.
- Key risks include the ENTR-601-44 clinical hold, early-stage and uncertain clinical development, the unproven EEV platform, manufacturing and third-party dependency, competition, intellectual-property exposure, and potential need for dilutive or otherwise costly financing. No material litigation was reported; management concluded disclosure controls were effective.
Investor verification priorities
- Confirm receipt and timing of the $75.0 million Vertex milestone and the relationship between the milestone, $77.5 million collaboration receivable, and revenue recognized.
- Track FDA interactions and progress toward lifting the ENTR-601-44 U.S. clinical hold, alongside UK Phase 1 data and planned regulatory filings.
- Assess whether the Q2 2026 cash-runway estimate remains supported by spending, Vertex research reimbursements, milestone receipts and development plans.
- Monitor R&D spending growth, working-capital movements, deferred-revenue recognition assumptions and any future ATM or other financing activity.
- Review subsequent Vertex VX-670 trial progress and any changes to collaboration scope, economics, or milestone expectations.
