Revenue grew 6.8%, mainly due to a higher average gross selling price. R&D rose substantially, including a $60 million upfront Stoke collaboration payment and a $10 million Neuren milestone, alongside higher pipeline development costs. SG&A fell as NUPLAZID commercial spending was optimized, partly offset by trofinetide launch preparation. Net loss widened 28.7% despite lower operating cash use. The company reported no debt in the balance sheet; liabilities included $62.0 million of operating lease obligations. Current assets were $507.9 million versus current liabilities of $125.6 million.

Q4 2022: Revenue was $136.5 million versus $130.8 million in Q4 2021. Net loss was $41.7 million versus $43.1 million, and loss per share was $0.26 versus $0.27.

Outlook, risks and notable items

  • Management expected available cash and investments to fund planned operations for at least 12 months from the filing, while noting possible future financing needs and continued losses over the next few years.
  • Trofinetide’s NDA had an FDA target action date of March 12, 2023. Approval and commercial success were uncertain; the pivotal trial showed meaningful gastrointestinal adverse events, including diarrhea in 80.6% and vomiting in 26.9% of treated participants.
  • Top-line results from the Phase 3 ADVANCE-2 schizophrenia study were expected in early 2024. ACP-204 was in Phase 1, with further Alzheimer’s disease psychosis studies planned subject to progress.
  • NUPLAZID sales remain central to the business. Four customers represented approximately 73% of FY2022 revenue. The product also has an ongoing FDA post-marketing commitment in predominantly frail and elderly patients.
  • Generic-patent litigation involving NUPLAZID was ongoing; a joint trial was scheduled for May 2023. Hetero settled and could launch its generic product July 27, 2038, subject to earlier-launch triggers. A securities class action was also pending. Acadia reported no loss accrual beyond legal fees and said it could not estimate potential losses.
  • Other material risks include regulatory and clinical-trial outcomes, outsourced manufacturing and supply concentration, reimbursement and drug-pricing changes, patent challenges, and lingering COVID-19 and geopolitical impacts.
