Changes versus the prior comparable period
- Net loss increased by $10.0 million, primarily reflecting higher operating expenses, partly offset by increased interest and other income.
- R&D expense rose 38% to $33.8 million. Management attributed the increase mainly to clinical activity, consultants, preclinical work and higher personnel costs as headcount grew.
- G&A expense rose 29% to $7.3 million, principally due to personnel and stock-based compensation, legal costs and external consultants.
- Operating cash use increased by $8.0 million to $32.0 million. Capital spending was lower: investing cash outflow was $0.04 million versus $1.8 million.
- Cash and restricted cash fell by $31.9 million during the quarter. The company had 35.9 million common shares outstanding at March 31, 2024, compared with 29.7 million a year earlier.
Programs, outlook and risks
- BMF-219 in type 2 diabetes: The company reported dose-escalation results in March 2024, including continued glycemic control reported at Week 26 after a four-week dosing period. The 200 mg with-food cohorts showed the largest reported placebo-adjusted HbA1c change, and the company said the drug was generally well tolerated in this study phase. Expansion cohorts were enrolling; initial 26-week data were expected in the second half of 2024.
- BMF-219 in type 1 diabetes: The Phase 2 COVALENT-112 trial is underway. In April 2024, after the reporting period, the company highlighted early activity and tolerability observations in its first two dosed patients. It expected 26-week follow-up data from the open-label portion in the second half of 2024.
- BMF-219 in oncology: COVALENT-101 reported early responses in a small group of patients with relapsed/refractory AML; the company cited two complete responses among seven patients evaluable for efficacy in reported data. Differentiation syndrome was reported in four participants, at Grade 3 or lower, and managed with treatment. Dose-escalation completion and selection of a recommended Phase 2 dose were expected in 2024. COVALENT-102 is evaluating BMF-219 in KRAS-mutated solid tumors, with dose-escalation completion and dose selection also expected in 2024.
- BMF-500: The Phase 1 COVALENT-103 trial for relapsed or refractory acute leukemia has dosed its first patient. Dose-escalation completion and selection of a recommended Phase 2 dose were expected in 2024.
- The company expects substantial and increasing operating losses as it advances clinical programs, and provides no specific revenue or earnings guidance. Clinical timelines and costs depend on enrollment, trial results, safety, regulatory decisions, manufacturing and third-party performance.
- Key risks include the need to raise capital, uncertainty of clinical and regulatory success, preliminary data from small patient populations, competition, reliance on external manufacturers and research organizations, and potential dilution or restrictive terms from future financing. Management said it was unaware of legal matters expected to have a material adverse effect; no material legal proceedings were reported.
