Changes versus the prior year
- Net loss widened by $13.1 million, reflecting higher operating expenses and lower interest income.
- R&D expense rose $2.3 million, primarily from increased payroll and headcount, partly offset by lower outsourced R&D activity.
- G&A expense increased $8.6 million, mainly due to payroll and stock compensation, market research, legal and professional fees, and insurance.
- Operating cash use increased by $7.4 million. Investing cash use was $11.2 million, including $14.8 million in property and equipment spending, principally for the ASTRA facility.
- Financing provided $118.0 million, primarily from the May 2020 public offering. Year-end cash increased $80.8 million.
Pipeline, outlook and material items
- B-VEC: Phase 3 GEM-3 began in July 2020 for dystrophic epidermolysis bullosa. Management expected enrollment completion in Q1 2021, top-line data in Q4 2021, and a U.S. BLA filing shortly thereafter; an EU application was also planned after the BLA.
- Other programs: KB105 was in Phase 1/2, with further patient enrollment planned in 2021. KB301 began a Phase 1 safety study in August 2020. Management expected to initiate a KB407 cystic-fibrosis trial in the first half of 2021 and file an IND for KB104 in the second half of 2021.
- The company expected no product revenue until 2022 at the earliest, assuming B-VEC approval on its contemplated schedule. It anticipated rising R&D and administrative costs and stated that profitability depends on successful development, approval and commercialization.
- Management said year-end cash and investments, together with subsequent financing, were expected to fund planned operations for at least 12 months from the filing date. After year-end, Krystal received $16.9 million net from ATM share sales and $135.0 million net from a February 2021 public offering; these amounts were not included in December 31, 2020 cash.
- ASTRA, a planned 150,000-square-foot manufacturing facility, was expected to be completed and validated in 2022. The company entered a $9.4 million purchase agreement in January 2021; the transaction’s completion was subject to customary conditions.
- COVID-19 had a minimal reported impact on the business and trials to date, but the company warned that restrictions, trial-site availability, staffing, supply chains and regulatory operations could cause delays.
- PeriphaGen alleged breach of contract and trade-secret misappropriation involving Krystal’s platform and product candidates. Krystal denied the claims and is defending the case; it could not estimate litigation costs, timing or potential damages.
Key investor verification points
- Check GEM-3 enrollment, results, safety and efficacy, and whether filing timelines or regulatory plans changed.
- Track trial progress and data for KB105 and KB301, and the planned KB407 and KB104 milestones.
- Compare cash burn and clinical/manufacturing spending with management’s stated runway; account for subsequent equity issuance and potential dilution under the $150 million ATM program.
- Verify ASTRA purchase and construction status, total costs, validation timeline and manufacturing readiness.
- Monitor the PeriphaGen litigation and any material developments in claims concerning ownership of the platform or trade secrets.
