Financial results and liquidity

MetricQ1 2024Q1 2023 / prior date
Total revenue$8.5 million$5.3 million
Operating loss$54.2 million$74.2 million
Net loss$65.6 million$77.2 million
Basic and diluted loss per share$1.36$1.63
Net cash used in operating activities$60.6 million$78.3 million
Cash, cash equivalents and restricted cash at quarter-end$246.2 million$157.0 million
Cash, cash equivalents and investment securities at quarter-end$555.7 million$617.9 million at December 31, 2023

Including restricted cash, total cash, cash equivalents and investment securities were $558.9 million at March 31, 2024. Current assets were $596.0 million and total liabilities were $623.3 million. Shareholders’ equity was $146.8 million, down from $207.7 million at year-end 2023.

Revenue comprised $1.2 million of HEMGENIX royalty revenue, $4.0 million of contract manufacturing revenue and $3.3 million of collaboration revenue. Revenue increased 59% year over year. Contract manufacturing costs were $9.1 million, versus $2.4 million; the company said the increase mainly reflected previously capitalized work-in-progress costs and a $1.8 million inventory write-down. These costs exceeded manufacturing revenue by about $5.1 million.

At quarter-end, long-term debt was $102.1 million and the royalty-financing liability was $405.4 million. The Hercules facility has $100.0 million principal outstanding, repayable in January 2027, with a variable interest rate subject to a 7.95% floor. Interest expense was $16.1 million, including $12.4 million recognized on the royalty financing; interest income was $6.5 million.

Changes versus the prior comparable period

  • Revenue rose by $3.2 million, mainly due to $1.2 million of newly recognized HEMGENIX royalties and collaboration revenue growth of $2.9 million; manufacturing revenue declined by $0.9 million.
  • R&D expense fell to $40.7 million from $60.8 million. The filing attributes the decrease to lower program and platform costs, reduced staffing and related expenses following the October 2023 reorganization, and the prior-year $10.0 million AMT-162 licensing-related expense.
  • SG&A declined to $13.9 million from $17.8 million, including lower professional fees and other costs.
  • Operating cash use improved by $17.7 million, while net loss narrowed by $11.6 million. Higher interest expense, including royalty-financing accretion, partly offset the improvement in operating loss.

Outlook, risks and unusual items

  • Management estimates that existing cash, cash equivalents and investment securities will fund operations into the second quarter of 2027, based on the current operating plan and assumptions. The estimate could prove wrong; additional funding may be needed to advance AMT-130 or other candidates into late-stage development.
  • Management planned to begin FDA discussions in Q2 2024 on AMT-130’s clinical and regulatory pathway, including interpretation of trial data and use of natural-history comparators. AMT-260, AMT-191 and AMT-162 were being advanced toward early-stage clinical trials.
  • Key risks include clinical and regulatory uncertainty—particularly for lead candidate AMT-130—trial delays or inconclusive results, gene-therapy safety and manufacturing challenges, dependence on CSL and other third parties, and the possibility that future financing is unavailable or dilutive.
  • The company has potential uniQure France milestone commitments of up to €190 million: €30 million associated with first dosing in an AMT-260 Phase I/II trial and €160 million tied to Phase III development and U.S./EU approvals. Timing and achievement are uncertain; up to 25% may be settled in shares.
  • The royalty-financing agreement provided $375 million upfront in exchange for specified HEMGENIX royalty rights. The purchaser is entitled to receive up to 1.85 times the upfront amount by June 2032, or up to 2.25 times through 2038 if the earlier cap is not met. A further $25 million milestone is conditional on HEMGENIX 2024 sales; specified terms may require a payment if the sales threshold is not met and a future CSL milestone is achieved.
  • The Hercules facility requires at least $30 million of unrestricted cash beginning April 1, 2024, and includes other covenants and collateral provisions. uniQure reported material covenant compliance at March 31, 2024. Fixed lease obligations were $50.7 million, including $8.4 million due within 12 months; future Hercules interest and financing fees were estimated at $44.2 million.
  • No legal proceedings or subsequent events were reported. Disclosure controls were considered effective; no material change in internal control over financial reporting was reported.

  • Whether the stated runway through Q2 2027 remains supported by updated cash-burn assumptions, clinical plans and reorganization savings.
  • HEMGENIX sales, royalty receipts and the terms governing the conditional $25 million royalty-financing milestone.
  • Manufacturing economics and inventory levels, including the causes and potential recurrence of manufacturing costs exceeding manufacturing revenue and inventory write-downs.
  • Timing and probability of AMT-260 milestones, potential settlement in shares, and the impact of royalty-financing accretion and Hercules debt service on future liquidity.
