Q1 revenue comprised $8.0 million for transferring future Beovu royalty rights to Novartis and $8.1 million of previously constrained royalties released after the related dispute was settled. These are not product-sales revenues and are not a recurring revenue base. The $8.0 million settlement payment was received on April 8, 2024; it was recorded as an $8.0 million receivable at quarter-end. Cost of revenue was $0.5 million, including the write-off of the remaining royalty-rights intangible asset.

Research and development expense rose 9% to $13.0 million, primarily due to higher PYX-201 clinical and manufacturing costs and PYX-106 trial costs. General and administrative expense fell 9% to $8.2 million, mainly due to lower professional and consulting fees. Other income was $2.3 million, including $1.6 million of interest and investment income and $0.8 million of sublease income.

At March 31, cash and cash equivalents were $28.0 million, short-term U.S. Treasury securities were $129.1 million, and restricted cash was $1.5 million. The company reported $157.0 million of cash, cash equivalents and short-term investments, $172.4 million of current assets, $14.1 million of current liabilities and $184.2 million of stockholders’ equity. Total liabilities were $36.1 million, including operating lease liabilities; the filing does not present a borrowing balance. Operating cash use was partly offset by $57.5 million of financing cash inflows. Cash, cash equivalents and restricted cash increased $18.3 million during the quarter.

Material changes and financing

  • Net loss narrowed by $16.0 million year over year, primarily because of the Novartis settlement-related revenue; operating expenses increased by $0.8 million.
  • The company raised $50.0 million gross in a February private placement through common shares and pre-funded warrants, and $10.8 million gross through its ATM program. Net financing proceeds were $57.5 million. The ATM had $107.9 million of remaining capacity at quarter-end.
  • Common shares outstanding increased from 44.8 million at December 31, 2023 to 58.8 million at March 31, 2024. An additional 1.6 million pre-funded warrant shares were outstanding, along with other equity awards and warrants.

Pipeline, outlook and risks

PYX-201: 42 patients had been dosed across eight Phase 1 cohorts; the company was studying 5.4–8 mg/kg and planned to dose 16 additional patients. It expected to report dose-finding results and next development milestones in fall 2024. PYX-106: 24 patients had been dosed in Phase 1; preliminary data were anticipated in the second half of 2024. PYX-107, acquired with Apexigen, has prior clinical experience; further development was still under portfolio evaluation. The company said it had paused preclinical development of PYX-203 and PYX-102 to prioritize spending.

Management stated that quarter-end cash, cash equivalents and short-term investments, together with the April Novartis payment, were expected to fund operations into the second half of 2026. The financial-statement note separately says quarter-end resources were expected to fund expenses and capital requirements for at least 12 months from issuance. Both estimates depend on assumptions and may change; management expects continued operating losses and may need additional financing.

Key risks include clinical-trial failure or delay, uncertain safety and efficacy, reliance on third-party manufacturers and clinical research organizations, competition, intellectual-property and licensing exposure, and the need for future capital, which could dilute shareholders. The filing reports no material legal proceedings. Management concluded disclosure controls were effective at a reasonable assurance level and reported no material change in internal control over financial reporting.

Investor verification priorities

  • Confirm the Novartis settlement accounting: $8.0 million cash received after quarter-end and $8.1 million of previously deferred royalties recognized in Q1.
  • Track clinical data and milestone timing for PYX-201 and PYX-106, including patient enrollment, safety, dose selection and any changes to stated reporting timelines.
  • Assess the assumptions behind the “second half of 2026” runway estimate, planned R&D spending and potential need for additional capital.
  • Monitor dilution from issued shares, pre-funded warrants, equity awards and remaining ATM capacity.
  • Follow management’s decision on PYX-107 and the status of paused PYX-203 and PYX-102 programs.
