Kymera Therapeutics KYMR is a clinical-stage biotechnology company focused on targeted protein degradation (TPD), an innovative approach that aims to eliminate disease-causing proteins that may be difficult to target with conventional therapies.
The company is developing an oral small-molecule degrader pipeline focused on immunological diseases, with the potential to address significant unmet medical needs.
KYMR is developing lead candidate KT-621, a once daily, oral degrader of STAT6, currently being evaluated in parallel phase IIb studies in atopic dermatitis (“AD”) and asthma. It completed enrollment in the BROADEN2 phase IIb study of KT-621 in moderate-to-severe AD nearly six months ahead of schedule. Top-line data are now expected by year-end 2026. Subject to regulatory discussions, phase III studies in atopic dermatitis are planned to begin by mid-2027.
In the phase I study in patients with AD, KT-621 demonstrated substantial STAT6 degradation in blood and skin, along with significant reductions in disease-relevant type 2 inflammatory biomarkers. The candidate also showed meaningful improvements in clinical measures and patient-reported outcomes in AD and comorbid conditions, including asthma and allergic rhinitis. Additionally, KT-621 was generally well tolerated and demonstrated a favorable safety profile.
KT-621 is also being investigated in the ongoing phase IIb BREADTH trial in moderate-to-severe eosinophilic asthma. Kymera expects top-line data in late 2027 and has initiated an open-label extension that permits eligible participants to receive KT-621 for up to 52 additional weeks.
The candidate has received Fast Track designation from the FDA for the treatment of moderate-to-severe AD and eosinophilic asthma.
Per KYMR, KT-621 is the first STAT6-directed drug to enter clinical evaluation and has the potential to address a significant unmet medical need among more than 140 million people worldwide living with type 2 inflammatory diseases.
The successful development of the candidate will be a significant boost for the company.
However, as its pipeline remains in clinical development, future growth will depend on clinical trial outcomes, regulatory progress and its ability to successfully commercialize its candidates.
Competition for KYMR’s in Protein Degrader Therapies
Arvinas ARVN is a clinical-stage biotechnology company pioneering PROTAC (PROteolysis TArgeting Chimera) protein degradation therapies.
Arvinas, in collaboration with Pfizer, developed the first FDA-approved PROTAC, a type of heterobifunctional protein degrader. The asset has since been licensed to Rigel Pharmaceuticals, Inc., which holds exclusive global rights for its development, manufacturing and commercialization.
Arvinas is advancing a clinical pipeline of investigational drugs targeting BCL6 (ARV-393) for non-Hodgkin lymphoma, LRRK2 (ARV-102) for neurodegenerative disorders, polyQ-AR (ARV-027) for spinal-bulbar muscular atrophy, HPK1 (ARV-6723) for solid tumors and KRAS G12D (ARV-806) for solid tumors. The company plans to seek an out-licensing partner for further clinical development of ARV-806.
ARVN recently announced that the first participant has been dosed in its phase I/II study of ARV-6723, an investigational, oral PROTAC designed to degrade hematopoietic progenitor kinase 1 (HPK1) in advanced solid tumors. ARV-6723 is Arvinas' first clinical candidate in IO and the first HPK1 PROTAC degrader to enter the clinic in the United States.
Nurix Therapeutics NRIX is another clinical-stage biopharmaceutical company developing targeted protein degradation therapies for cancer and autoimmune diseases. Nurix is advancing bexobrutideg, a BTK degrader co-developed with Roche, and NX-1607, a CBL-B inhibitor, for oncology. Its autoimmune pipeline features IRAK4 degraders developed in collaboration with Gilead and STAT6 degraders partnered with Sanofi.
NRIX is also developing early-stage degraders and degrader antibody conjugates through its proprietary research platform and partnerships with Gilead, Sanofi and Pfizer.
