Research and development expense rose 121% to $30.1 million, primarily reflecting greater clinical-trial and trial-material costs, personnel costs, severance, and a $5.6 million noncash expense from equity-award modifications. General and administrative expense increased 53% to $14.1 million, including higher stock compensation and personnel costs. Sales and marketing expense fell to near zero after the company exited YUTIQ promotion. Interest and other income was $4.0 million, compared with $1.2 million; no interest expense was recorded in Q1 2024.
At quarter-end, total assets were $329.2 million, total liabilities $79.3 million, and stockholders’ equity $249.9 million. Current assets were $317.7 million and current liabilities $62.6 million. The company reported no outstanding debt; it repaid and terminated its Silicon Valley Bank loan in May 2023. Operating lease liabilities totaled $5.7 million. Q1 operating cash outflow exceeded net loss partly due to working-capital movements, including $8.8 million of deferred revenue recognized against the Alimera arrangement. Stock-based compensation was $12.7 million.
Material changes and notable items
- Revenue mix shifted substantially: product sales declined after YUTIQ rights transferred to Alimera, while $10.4 million of Q1 license and collaboration revenue came from recognizing deferred revenue under the Alimera product-rights and supply arrangement.
- Alimera paid a $75 million upfront amount in 2023 and is to make four quarterly payments totaling $7.5 million during 2024. At March 31, 2024, $37.0 million of the related deferred revenue was classified as current.
- Research and development spending increased as the company advanced DURAVYU trials in wet AMD, NPDR and DME. The company expects to initiate pivotal Phase 3 wet AMD trials in the second half of 2024.
- Common shares outstanding were 49.9 million at March 31, versus 49.0 million at year-end; the filing reports 52.1 million shares outstanding on May 2. Equity awards and potential dilution are also significant.
Outlook, risks and contingencies
- Management said the $299.3 million of cash, cash equivalents and marketable securities was expected to fund the current operating plan through topline data from planned Phase 3 wet AMD trials, into 2026. The company expects substantial continuing losses and may need additional capital; trial results, spending, financing availability and collaboration income could alter the runway.
- The filing also states that the cash position was expected to fund current and planned operations for at least the next twelve months. These statements are management estimates, not guarantees.
- In May 2024, after quarter-end, the company reported that the Phase 2 PAVIA trial of DURAVYU in NPDR showed biological effect and a favorable safety and tolerability profile but did not meet its prespecified primary endpoint. It said it would review full 12-month data before updating its plans. This subsequent development is not a Q1 operating result.
- The FDA issued a Form 483 following a February 2024 inspection of the Watertown facility and classified the facility as Official Action Indicated (OAI). The company said it was addressing the observations; the filing warns that unresolved issues could affect YUTIQ manufacturing or lead to enforcement action.
- The company is cooperating with a U.S. Department of Justice investigation following an August 2022 subpoena concerning sales, marketing and promotional practices, including DEXYCU. It cannot predict the matter’s scope, duration or outcome, or whether it will have a material effect.
- Other risks include clinical-trial outcomes and timing, reliance on third-party research and manufacturing providers, manufacturing and supply constraints, intellectual-property protection, and the need for future funding. The filing notes potential U.S. restrictions affecting WuXi AppTec, a provider supporting EYP-2301 development.
Investor verification priorities
- Track FDA feedback and the timing and execution of the planned Phase 3 wet AMD trials, as well as the full PAVIA data and the company’s NPDR plans.
- Monitor corrective actions and FDA follow-up regarding the Watertown OAI classification and any implications for YUTIQ supply to Alimera.
- Review the DOJ investigation for developments, potential consequences and any changes in the company’s disclosures.
- Assess cash burn, the timing of Alimera payments and deferred-revenue recognition, and whether management’s stated runway remains supported by trial and operating costs.
- Verify the evolving share count, equity-award dilution and the effect of the substantial stock-based compensation expense.
