- 2021 revenue and profit comparisons reflect the one-time $462.4 million CSL Behring license-sale revenue recognized in May 2021. 2022 revenue was mainly collaboration services; no license revenue was recognized.
- Six-month R&D expense rose $26.4 million, reflecting increased spending on AMT-130 and AMT-260, personnel, facilities and manufacturing supplies. SG&A fell $6.8 million, primarily because licensing-related advisory and professional fees were lower.
- Six-month net loss included $28.0 million in net foreign-currency gains, partly offset by $5.2 million interest expense. These currency gains materially supported reported results.
- At June 30, cash and cash equivalents were $500.5 million; cash, cash equivalents and restricted cash totaled $503.7 million, versus $559.4 million at December 31, 2021. Total assets were $701.2 million and total liabilities $214.7 million.
- Debt principal outstanding was $100.0 million. The facility matures in December 2025; principal repayment is scheduled between December 2024 and December 2025, subject to specified conditions. The company reported compliance with its debt covenants.
- Management said existing cash was expected to fund operations into the first half of 2025, assuming achievement of $175.0 million of first-commercial-sales milestones under the CSL agreement. This runway estimate is conditional.
Business developments, outlook and risks
- AMT-130: In June, uniQure reported low-dose cohort data at 12 months: no serious adverse events considered related to treatment, and mean CSF mutant huntingtin levels fell 53.8% in four treated patients with evaluable data. These are small-cohort interim findings, not proof of efficacy.
- In July 2022, after quarter-end, three of 14 patients treated at the higher dose in the European study experienced serious reactions potentially attributable to AMT-130. The independent safety board recommended pausing further high-dose enrollment and dosing pending a comprehensive review. The company said it would follow that recommendation; timing was expected in Q3 or early Q4 2022.
- Hemophilia B / CSL Behring: CSL submitted U.S. and EU marketing applications in March and April and uniQure collected the related $55.0 million milestone. The FDA accepted the BLA for Priority Review. After quarter-end, the EMA’s advanced-therapy committee indicated that it could not meet the accelerated timetable and would use a standard review. The company also reported that its Lexington site could receive GMP certification for commercial supply following an inspection.
- Management anticipated higher expenses as it advanced clinical and preclinical programs, expanded manufacturing capabilities and pursued platform development. It noted that research and development costs may vary with trial activity, manufacturing campaigns and regulatory work.
- Key risks include clinical safety or efficacy setbacks, regulatory delays, manufacturing and supply constraints, dependence on collaborators and third parties, the need for future financing, and uncertainty about commercialization, reimbursement and market acceptance. COVID-related supply lead times had increased, though management said no material timeline impact had occurred to date.
- Pfizer’s patent challenges to two uniQure patents were ongoing; substantive briefing was completed in July 2022 and a PTAB hearing was planned for August. The filing reported no new material legal proceedings beyond previously disclosed matters.
Investor verification checklist
- Confirm subsequent AMT-130 safety-review findings, any regulatory actions and the status of high-dose enrollment and dosing.
- Track FDA and EMA review timelines for etranacogene dezaparvovec, including the effect of the EU move to standard review and any commercial milestones or royalties.
- Assess cash use against the stated first-half 2025 runway assumption, especially the timing and achievement of the $175.0 million in first-commercial-sales milestones.
- Review the debt covenants, future principal and interest payments, and potential Corlieve milestone obligations; up to $210.3 million of nominal Corlieve milestones were described, subject to development and regulatory achievements.
- Monitor R&D spending, foreign-exchange effects on reported earnings, and the outcome of the Pfizer patent proceedings.
