Research and development costs rose mainly from external discovery and preclinical work and increased research headcount. General and administrative costs increased with staffing, professional services and public-company insurance and other costs. No meaningful revenue-based margin comparison is available. The company reported no debt; total liabilities were $39.3 million, including $31.9 million of operating lease obligations. Stockholders’ equity was $277.4 million.

Cash, cash equivalents and restricted cash fell by $202.5 million during the quarter, while the company used $181.5 million in investing activities, primarily to purchase marketable securities. This largely reflects reallocating cash into investments, rather than operating burn alone. Marketable securities had $1.5 million of unrealized losses, recorded in other comprehensive loss; the company attributed these principally to interest-rate changes and reported no impairment.

Material changes and unusual items

  • Operating expenses increased $13.8 million year over year, and net loss increased $13.3 million.
  • Entrada adopted lease-accounting standard ASC 842 on January 1, 2022, recognizing operating lease assets and liabilities. Adoption had no impact on results of operations or operating cash flows.
  • In March 2022, Entrada signed a lease for approximately 81,442 square feet of additional Boston office and laboratory space, intended to become its consolidated headquarters. The lease had not commenced by quarter-end and was therefore not included in reported lease liabilities. It has an approximately 10-year term; initial fixed rent is $0.5 million per month for a 12-month period, followed by annual increases. The landlord allowance for initial build-out is up to $19.5 million. A $3.9 million cash-collateralized letter of credit is reported as restricted cash.
  • Common stock and additional paid-in capital reflect the company’s post-IPO capital structure. The 2021 IPO generated approximately $190.7 million of net proceeds; no material change to the planned use of proceeds was reported.

Outlook, commentary and principal risks

  • Management expects cash, cash equivalents and marketable securities at March 31, 2022 to fund operating expenses and capital requirements into the second half of 2024. The financial-statement liquidity note separately states the resources are expected to cover at least the next 12 months. Both estimates depend on assumptions and spending could exceed expectations.
  • Management expects ongoing operating losses and negative operating cash flows, with research and development spending expected to increase as programs advance. Further financing will be needed to support operations and strategy; additional funding may be unavailable or costly, dilutive, or require surrendering rights.
  • At the time of filing, Entrada planned to submit an IND for ENTR-601-44, its DMD exon 44 program, in Q4 2022, and an IND for ENTR-701, its DM1 program, in 2023. The filing reports that on May 11, 2022, after quarter-end, the company announced preclinical ENTR-601-44 exon-skipping results in non-human primates through 12 weeks after one infusion and selected ENTR-701 as its DM1 clinical candidate based on preclinical findings. These are preclinical results, not clinical evidence.
  • Key risks include the unproven EEV platform, lack of human clinical data, uncertain translation of animal results, regulatory and manufacturing challenges, dependence on CROs and manufacturers, competition, intellectual-property risks, and the need for additional capital. COVID-19 had caused temporary operational reductions that the company said had normalized; it reported no significant pandemic-related impairment or financial impact to date.
  • Management reported no material litigation at quarter-end and concluded disclosure controls were effective at the reasonable-assurance level. No material change in internal control over financial reporting was reported for the quarter.

Important facts for investors to verify

  • Whether the IND timelines for ENTR-601-44 and ENTR-701 were met, and whether subsequent regulatory or preclinical findings changed program plans.
  • Whether the reported animal-model results are replicated and support safe, effective clinical development; no candidate had entered clinical trials as of the filing.
  • Actual quarterly cash burn, investment maturities and liquidity runway compared with management’s second-half-2024 estimate.
  • The commencement, total costs and cash-flow effects of the new headquarters lease, including build-out spending and the restricted-cash letter of credit.
  • Future operating-expense growth, financing needs and potential dilution, given the absence of revenue and expected continuing losses.
